Abstract : Identifying functional protein sites and understanding which and how genetic variations alter them are central questions for bioengineering, medicine, and fundamental biology. I will present some of our recent developments for assessing the impact of sequence variations on how protein function and interact with one another. I will focus on single-point mutations and alternative splicing-induced changes. Our approaches harness the vast reservoir of protein sequence and structural information accessible in public databases. Their efficiency and robustness make them amenable to comprehensive proteome-wide investigations.
