Philippe Cluzel Measuring multi-gene transcriptional activity to characterize the assembly of the bacterial motor in individual E coli

Over the last decade time-lapse microscopy has allowed for high-precision measurements of gene transcriptional activity in individual bacteria. However, some key properties of transcriptional systems have not been measurable because they are associated with signals that are either too fast or too weak to be measured across successive cell divisions. I will introduce new technical improvements that allow us to characterize the transcriptional dynamics that underlie the assembly of the bacterial motor in single bacteria. We have found that large classes of the motor genes are pulsating synchronously. Our measurements suggest that such pulses are not generated directly at the transcription level, but by an autonomous mechanism.
